Introduction

Testing antibacterial activity is carried out on two types of harmful bacteria, namely staphylococcus aureus and pseudomonas aeruginosa. Hydrogel preparations are made with variations in the concentration of kirinyu leaf extract, namely F1 (3%), F2 (5%), and F3 (7%), and a combination of hydrogels with 1% collagen, namely F1K (3% + 1%), F2K (5% + 1%), and F3K (7% + 1%). As a control, collagen hydrogen preparations are also made without kirinyu leaf extract (F0K) and basic hydrogel (F0).
Evaluation of antibacterial activity and dosage stability
Evaluation carried out includes testing antibacterial activity and stability of hydrogel preparations. The results of characterization showed several important parameters, such as water content (7.32%), water soluble levels (24.97%), extract levels soluble in ethanol (17.45%), as well as total ash content and insoluble ash content in acids each is 7.78% and 1.09%. Testing antibacterial activity indicates that the ethanol extract of kirinyu leaves is effective in inhibiting the growth of the two pathogenic bacteria. Hydrogel preparations F1, F2, F3, F1K, F2K, and F3K show good ability to inhibit bacterial growth, while F0K and F0 do not show the same effectiveness.
Excision Wound Healing
Testing excision wound healing is carried out on mice treated with hydrogel preparations that have met the stability standards. The results of the analysis using One Way Anova showed that hydrogel preparations F1, F2, F1K, F2K, and F0K were given significant results in reducing the diameter of the wound compared to the negative control group (F0). Interestingly, there is no significant difference between combination hydrogel preparations and positive controls (Octenilin Gel).
Of all the preparations tested, F2K hydrogels show the greatest potential in wound healing with healing time reaching 18 days. Its antibacterial activity is also higher (11.20 mm) compared to Octenilin Gel (10.33 mm).
Conclusion
Based on the results of this study, it can be concluded that hydrogel preparations containing ethanol extracts of kirinyu and collagen leaves have excellent potential as an alternative to healing excision wounds. This combination not only shows effectiveness in inhibiting bacterial growth, but also accelerates the wound healing process. It is hoped that these findings can be a basis for the development of wound healing products that are safer and effective using natural ingredients.
